(1S)-1-Phenyl-1,2,3,4-tetrahydroisoquinoline
(S)-1-phenyl-1,2,3,4-tetrahydroisoquinoline can be used to prepare solifenacin. Solifenacin succinate, CAS No. 242478-38-2, chemical name 1-azabicyclo[2.2.2]octan-8-yl-(1S)-1-phenyl-3,4-dihydro-1H-isoquinolin e-2-carboxylate succinate, is prepared by condensing (S)-1-phenyl-1,2,3,4-tetrahydroisoquinoline (I) with triphosgene under organic base (weak base) conditions to obtain (S)-1-phenyl-3,4-dihydro-1H-isoquinoline-2-carbonyl chloride (II). Compound II can be directly heated in the presence of an organic base without separation through a one-pot method, and condensed with (R)-3-quinuclidine to obtain solifenacin (III), which is then salified with succinic acid to obtain solifenacin succinate (IV).
